目的研究消退过程中豚鼠眨眼条件反应特征参数的变化规律。方法 27只雄性12~16周龄豚鼠(体质量450~600 g),按随机抽签法分为:1延迟性眨眼条件反射组(n=13);2痕迹性眨眼条件反射组(n=14)。使用2 k Hz的正弦波纯音和医用氧气流分别作为条件刺激和非条件刺激,配对训练豚鼠建立延迟性和痕迹性眨眼条件反射。而后单独给予纯音条件刺激,使豚鼠已经建立的眨眼条件反射消退。高精度张力换能器记录豚鼠左侧的眼轮匝肌活动,Matlab程序分析眨眼运动行为数据。结果与痕迹组比较,延迟组动物条件眨眼反应习得率和条件眨眼反应峰幅度均显著增高(P〈0.05),但两组条件眨眼反应的消退速率没有显著区别(P〉0.05)。此外,延迟组豚鼠反射消退较痕迹组自发恢复更明显(P〈0.05)。结论延迟性和痕迹性眨眼条件反应的消退具有不同的行为学特征。
Objective To investigate the behavioral characteristics of eyeblink responses during the extinction of delay and trace eyeblink conditionings in guinea pigs. Methods A total of 27 male guinea pigs( 12- 16 weeks-old,450- 600 g) were randomly assigned into 2 groups,delay eyeblink conditioning group( n = 13),and trace eyeblink conditioning group( n = 14). Pure tone conditioned stimulus( CS) of 2 k Hz was paired with corneal oxygen-puff unconditioned stimulus( US) to acquire delay and trace conditioned eyeblink responses( CR). The guinea pigs received CS alone during the extinction. A high-resolution springreturn potentiometer was used to monitor the activities of left orbicularis oculi muscles. Self-developed software written in the Matlab environment was used to quantitatively analyze the behavioral data. Results Compared with the trace-conditioned guinea pigs,the delay-conditioned guinea pigs acquired faster CRs( P〈0. 05) and higher peak amplitude( P〈0. 05). Nevertheless,there was no significant difference in the CR extinction rate between the 2 groups( P〉0. 05). In addition,more obvious spontaneous recovery of the extinguished CRs was observed in the delay-conditioned guinea pigs( P〈0. 05) as compared to the trace-conditioned guinea pigs.Conclusion The behavioral characteristics of delay CR extinction are different from those of trace CR extinction.
