目的:探讨大鼠局灶性脑缺血损伤后Cdh1 mRNA水平的变化及其在不同类型神经细胞中的分布变化.方法:雄性成年SD大鼠30只,线栓法建立右侧大脑中动脉缺血模型,分别于缺血前和缺血再灌注后1、3、7 d采用实时荧光定量PCR技术检测缺血侧和非缺血侧脑组织中Cdh1 mRNA的水平 缺血再灌注后7 d免疫荧光双标法检测Cdh1在神经元和星形胶质细胞中的分布情况.结果:脑缺血前,两侧脑组织中Cdh1 mRNA的水平差异无统计学意义.缺血再灌注后1、3、7 d,缺血侧脑组织Cdh1 mRNA水平均低于非缺血侧脑组织(P<0.05) 缺血再灌注后7 d,与非缺血侧相比,缺血侧神经元明显减少,星形胶质细胞增多 在非缺血侧Cdh1主要表达于神经元,星形胶质细胞中表达较少,在缺血侧Cdh1在神经元及激活的星形胶质细胞中均有表达.结论:大鼠局灶性脑缺血损伤后,缺血侧Cdh1 mRNA的水平较非缺血侧降低,且Cdh1在神经元和星形胶质细胞中分布情况发生变化.
Objective: To investigate the changes of transcription of Cdhl mRNA and distribution of Cdh1 protein in rats after focal cerebral isehemia. Methods.. Thirty adult male Spra- gue-Dawley rats were studied. Middle cerebral artery occlusion (MCAO) was induced by inser- ting a nylon thread with rounded tip at bifurcation of the right common carotid artery into the in- ternal carotid artery. At pre- and 1, 3 and 7 d post-reperfusion, the transcription level of Cdhl mRNA in ischemic hemisphere and the non-ischemic hemisphere brain tissues was determined by quantitative PCR. On the 7th day after reperfusion, doubledabeled immunofluorescence assay was used to observe the distribution of Cdh1 protein in different neural cells. Results: At differ- ent time points after ischemia-reperfusion, the transcription level of Cdhl mRNA in ischemic tis- sue was significantly decreased than that in non-ischemic tissue (P〈 0. 05). Seven days after reperfusion,the number of neurons was decreased and astrocytes increased in the ischemic brain tissue relative to that in the non-isehemic brain tissue. The Cdh1 protein was highly expressed in both the ischemic and non-ischemic neurons. Cdhl-positive astrocytes were observed in the ische- mic brain tissue, but scarcely seen in the non-ischemic brain tissue. Conclusion: The transcrip- tion of Cdhl mRNA in the isehemic brain tissue was down-regulated and the cell distribution of Cdh1 protein changed after focal cerebral ischemia.
