目的:研究红活麻总香豆素的制备工艺。方法:通过正交设计试验选择红活麻总香豆素的最佳提取条件,采用大孔吸附树脂对红活麻总香豆素进行纯化,紫外分光光度法测定总香豆素的含量。结果:红活麻总香豆素的最佳提取工艺为以4倍体积70%乙醇,提取3次,每次1.5 h。HPD300大孔树脂分离纯化红活麻总香豆素效果最佳,其工艺条件为:上样浓度0.4 g生药/mL,上样体积为30 mL,流速为2 BV/h,先用4 BV的蒸馏水除杂,再用4BV的70%乙醇洗脱;得率可达52%以上。结论:该方法适合于制备红活麻总香豆素,工艺的稳定性和重复性较好。
Objective:To investigate the preparation process of total coumarins from Laportea bulbifera. Methods:The optimum con- dition for the extraction of total coumarins was studied using the orthogonal test, and the extracts was purled with different kinds of macroporous resins. The content of total coumarins was detected by ultraviolet spcctrophotomctry. Results: The optimum extraction process was as follows: three-time circumfiuence with 4 times volume of 75% ethanol for 1.5 h each time. HPD300 macroporous resin was the optimum one for the separation and purification of total eoumarins. The extraction condition was loading 30mL of 0. 4 g/mL the crude drug ,flowing at 2BV/h, then edulcorating with 4 times volume of distilled water and eluting with the same volume of 75% ethanol. The yield rate was more than 52%. Conclusion:This method is suitable for the preparation of total coumarins in Laportea bulbifera, and the process is stable and repeatable.
