目的:探讨左、右归丸增加去势大鼠阴道固有层血管数量的机制。方法:将雌性育龄期大鼠随机分为正常组、假手术组、去势模型组、去势倍美力组、去势左、右归丸组。正常组、假手术组、去势模型组:每日生理盐水灌胃;去势倍美力组:以62.5μg/100 g倍美力灌胃;去势左、右归丸组:按成人用量的15倍灌胃。连续给药12周。采用免疫组化法阴道组织进行病理切片制作,通过Mias-2000图像分析系统检测阴道黏膜雌激素受体α(ERα)及雌激素受体β(ERβ)蛋白表达水平,并测量阴道黏膜厚度、上皮层数、皱襞数和阴道固有层血管数;采用荧光定量PCR技术检测阴道组织ERαmRNA表达水平。结果:与假手术组、正常组比较,去势模型组大鼠阴道黏膜厚度、阴道皱襞数、阴道固有层血管数明显降低(P〈0.05);与去势模型组比较,去势倍美力组大鼠阴道黏膜厚度、阴道皱襞数、阴道固有层血管数明显增加(P〈0.05),去势左、右归丸组大鼠阴道皱襞数、阴道固有层血管数均明显升高(P〈0.05);与假手术组、正常组比较,去势模型组大鼠阴道ERα与ERβ明显减少(P〈0.05);与去势模型组比较,去势倍美力组、去势左丸组大鼠阴道ERα明显增加(P〈0.05),去势倍美力组、去势左、右归丸组大鼠阴道ERβ明显增加(P〈0.05);假手术组、正常组、倍美力组、左、右归丸组大鼠阴道组织ERαmRNA的表达量分别为模型组的1.83、1.54、1.53、1.88、1.36倍。结论:左归丸通过促进去势雌性大鼠阴道ERα、ERβ蛋白及mRNA表达、右归丸通过促进去势雌性大鼠阴道ERβ蛋白及mRNA表达而促进去势雌性大鼠阴道固有层血管数量。
Objective:This study was to investigate the mechanism of increasing vaginal ER in ovariectomized rats by Zuogui Wan (ZGW) and Yougui Wan (YGW). Methods:Female sexual matured rats were randomly divided into 6 groups: normal group, sham-operated group,ovariectomized group, ovariectomized Premarin group, ovariectomized ZGW and YGW group. Normal group ,sham-operated group ,model group were fed by saline. Rats in Premarin group were fed by Premarin(62.5 μg/100g) ;rats from ZGW and YGW group were fed by the formula respectively in dosage of 15 folds of adults. All rats were fed for 12 weeks. The thickness of vaginal mucosa, covering epithelium, vaginal folds and vessel numbers of lamina propria were measured by Mias - 2000 image analysis system. The expression level of ERa and ERβ in the uterus and vagina was measured by immunohistochemis- try. The expression of vaginal ERa mRNA was measure by fluorescent quantitation PCR. Results : Compared with sham-operated and normal groups, the thickness of vaginal mucosa,vaginal folds and vessel numbers of lamina propria in model group were de- creased significantly ( P 〈 0. 05 ). Compared with model group, the thickness of vaginal mucosa, vaginal folds and vessel numbers of lamina propria in Premarin group were increased significantly ( P 〈 0. 05 ) , the thickness of vaginal folds and vessel nmnbers of lamina propria in ZGW and YGW group were increased significantly ( P 〈 0. 05 ). Compared with sham - operated and normal groups, the vaginal ERα and ERβ in model group were decreased significantly (P 〈 0.05). Compared with model groups,the va- ginal ERα in Premarin and ZGW groups were increased significantly (P 〈 0. 05 ) , the vaginal ERβ in Premarin, ZGW and YGW groups were increased significantly (P 〈 0. 05 ).
