目的研究小叶黑柴胡多糖(BPs)对巨噬细胞免疫功能的影响。方法收集KM小鼠腹腔巨噬细胞,用不同质量浓度的BPs溶液(10、100、200mg·L^-1)刺激,检测巨噬细胞吞噬E.coli、趋化和分泌一氧化氮(N0)的功能。结果BPs各个浓度组均能显著促进巨噬细胞吞噬E.coli和趋化作用(P〈0.001),但对巨噬细胞分泌NO无明显影响;BPs100、200mg·L^-1可明显抑制脂多糖诱导的巨噬细胞分泌NO(P〈0.05)。结论BPs可增强巨噬细胞免疫功能,但能抑制脂多糖诱导的炎症介质分泌。
AIM To investigate the effects of polysaccharides from Bupleunan smithii var. parvifolium (BPs) on the immunologic functions of macrophages. METHODS Murine peritoneal macrophages were obtained and treated with different concentrations of BPs ( 10, 100,200 mg·L^-1 ) to evaluate their phagocytic capacity towards Escherichia coli, chemotaxis and the ability of nitric oxide(NO) secretion. RESULTS BPs, at the dosage used in this study, obviously promoted phagocytosis towards Escherichia coli and chemotaxis ( P 〈 0. 001 ), but had no effect on NO production of macrophages. BPs 100 and 200 mg·L^-1 significantly suppressed lipopolysaccharide-induced NO secretion of macrophages ( P 〈 0.05). CONCLUSION BPs could enhance the immunologic function of macrophage, but inhibit the secretion of lipopolysaecharide-induced inflammatory mediator.
