目的:观察养心通脉片(YTT)对冠心病大鼠痰瘀痹阻证脂质代谢SREBP2通路基因m RNA的影响。方法:采用维生素D3和异丙肾上腺素复合高脂饲养建立大鼠心肌缺血痰瘀痹阻模型,随机分为3组。YTT组服YTT,绞股蓝总苷片(MET)组服MET,模型组不给药,另设正常大鼠为正常组。实验结束后检测血脂、血液流变学、用HE染色观察冠状动脉组织学变化,用逆转录聚合酶链反应(RT-PCR)法检测脂质代谢通路基因m RNA表达的变化。结果:YTT对冠心病大鼠有明显的降脂效果,能改善血液流变性,并能消退斑块,维持冠状动脉组织结构及功能,改善脂质代谢SREBP2通路基因m RNA的异常表达,使之趋于正常。结论:YTT具有良好的改善冠心病大鼠痰瘀病理的作用,它可能是通过调整脂质代谢SREBP2通路基因m RNA的表达来实现的。
Objective: To observe the effects of Yangxin Tongmai Tablet(YTT) on relevant genes expression of SREBP2 lipid metabolism pathways in CHD rats with phlegm and blood stasis obstruction syndrome. Methods: The rat model of myocardial ischemia with phlegm and blood stasis obstruction syndrome was established with the high-fat feed mixing with vitamin D3 and isoprenaline. The rats were randomly divided into the control group, model group, YTT group, and gynostemma total glycosides(MET). The YTT group was given the drug administration of YTT, and the MET group was given the treatment of MET. The blood lipid and hemorheology were detected, and the morphological changes in coronary artery were observed with immunohistochemistry staining. Meanwhile, the relevant genes expression of SREBP2 lipid metabolism pathways were detected by reverse transcription polymerase chain reaction(RT-PCR) method. Results: YTT significantly decreased the blood lipid, improved hemorheolgy, removed hyperplastic plaque of arterial tissue, maintained the structure and function of coronary artery, and regulated the abnormal genes expression in SREBP2 lipid metabolism pathways. Conclusion: YTT could effectively improve the CHD rats with phlegm and blood stasis obstruction syndrome by regulating the relevant genes expression in the SREBP2 lipid metabolism pathways.
