采用LPS刺激巨噬细胞RAW264.7建立体外的炎症模型,探讨灰毡毛忍冬不同极性萃取部位抗炎活性和作用机制.用MTT法检测灰毡毛忍冬不同极性萃取部位对RAW264.7细胞的毒性作用;ELISA法检测灰毡毛忍冬不同极性萃取部位对IL-6和TNF-α表达情况的影响;结果显示:灰毡毛忍冬不同极性萃取部位在一定的剂量范围内均能抑制炎症因子IL-6和TNF-α表达,其中石油醚部位(100 μg/mL)对IL-6表达量以及水部位(100μg/mL)对TNF-α表达量没有显著性的抑制作用,而乙酸乙酯部位对IL-6表达量和TNF-α表达量的抑制作用都最为明显.灰毡毛忍冬不同极性萃取部位都有抗炎活性,并且各个部位均有浓度依赖性.
To establish in vitro Inflammation model by using LPS to stimulate RAW264.7 macrophages,MTT method was adopted to detect the toxicity of extracts with different polarities from Lonicera macranthoides on RAW264.7 cells.Effects of extracts with different polarities on the expression of IL-6 and TNF-α were detected by ELISA method.Anti-inflammatory activity and mechanism of different polar extraction parts from L.macranthoides was discussed.All the extraction parts with different polarities from L.macranthoides could inhibit the expression of IL-6 and TNF-α.Inhibitory effects of the petroleum ether fraction (100 μg/mL) on the IL-6 expression and water extracts (100 μg/mL) on the TNF-α expression were not significant,while inhibitory effect of the ethyl acetate fraction on the IL-6 and TNF-α expression was the most obvious (P < 0.01).All extraction parts with different polarities from L.macranthoides have anti-inflammatory activity and are concentration dependent.
