目的观察弓形虫复合黏膜疫苗滴鼻免疫BALB/c小鼠后诱导的血清IgG、粪便和鼻咽冲洗液中IgA特异性抗体动态,探讨其抗弓形虫感染作用机制。方法96只BALB/c小鼠随机分为实验组和对照组,实验组以弓形虫复合黏膜疫苗(20μl/只)滴鼻免疫,对照组以PBS滴鼻。分别于滴鼻2次(间隔2周)后第1、2.3、4、6、8、10和12周处死小鼠,收集血清、粪便及鼻咽冲洗液,采用ELISA法检测血清IgG以及粪便和鼻咽冲洗液中IgA抗体。结果实验组小鼠血清IgG、粪便及鼻咽冲洗液中IgA均高于对照组,血清IgG水平第3周达高峰,第1~4、6和8周显著高于对照组;粪便IgA抗体第2周达高峰,第2、3和4周显著高于对照组;鼻咽冲洗液中IgA第1周即达高峰,之后逐渐降低,至第12周仍显著高于对照组。结论弓形虫复合黏膜疫苗滴鼻免疫BALB/c小鼠可诱导高水平的特异性抗体,且可持续较长时间。
Objective To observe the dynamics of IgG in sera as well as IgA in fecal and nasal washing of mice immunized with mucosal complex vaccine against Toxoplasma gondii by intranasal drip and explore the mechanism of the vaccine against infection with Toxoplasma gondii. Methods Ninty-six BALB/c mice were randomly divided into test and control groups and immunized with 2 doses of mucosal complex vaccine against Toxoplasma gondii and PBS respectively,at an interval of 2 weeks. Kill the mice on weeks 1, 2,3,4,6,8,10 and 12 after the 2nd dose respectively and determine the IgG in sera as well as IgA in fecal and nasal washing by ELISA. Results The IgG level in sera of mice in test group reached a peak value at week 3 and was significantly higher than that in control groups during weeks 1-4,6 and 8. The IgA level in fecal of mice in test group reached a peak value at week 2 and was significantly higher than thai in control group during weeks 2-4. However,the IgA level in nasal washing of mice in test group reached a peak value at week 1 then decreased gradually,but was still significantly higher than that in control group until week 12. Conclusion The immunization with mucosal complex vaccine against Toxoplasma gondii by intranasal drip induced high and persistent specific antibody level in mice.
