目的探讨温润通脉方治疗大鼠心律失常的作用及相关机制,为其临床的进一步应用提供实验依据。方法 SD雄性大鼠32只,随机分为正常组、模型组、西药组(即普萘洛尔组)、复方组(即温润通脉方组),每组8只,正常组和模型组采用生理盐水灌胃(2 m L/200 g),西药组采用普萘洛尔溶液灌胃(0.54 mg/200 g),复方组采用温润通脉方灌胃(1.17 g生药/200 g),药物干预7 d后,制备乌头碱诱发大鼠心律失常模型,颈动脉采血及取心脏组织,检测血清及心脏组织SOD、MDA、NO含量。结果复方组能提高血清SOD含量,与模型组相比差异有统计学意义(P〈0.05)。复方组能降低组织NO含量,和模型组相比差异有统计学意义(P〈0.01)。结论复方组即温润通脉方能增加SOD活力,提高NO含量,表明其可能通过清除氧自由基,提高抗氧化酶活性及抗脂质过氧化能力等途径,发挥减轻心律失常发生的作用。
Objective To investigate the effects of Wenrun Tongmai prescription on treatment of arrhythmia in rats and its related mechanism, and provide experimental basis for the further clinical application. Methods 32 SD male rats were randomly divided into the normal group, model group, the western medicine group(propranolol group), the TCM compound group(Wenrun Tongmai prescription group), 8 rats in each group, the normal group and model group by normal saline lavage(2 m L/200 g), western medicine group by propranolol solution lavage(0.54 mg/200 g), compound group by Wenrun Tongmai prescription lavage(1.17 g drugs/200 g). After 7 days of drug intervention, the arrhythmia model rats were induced by aconitine, and then acquire carotid artery blood and cardiac tissue to detect the content of SOD, MDA and NO.Results The serum SOD levels in TCM compound group were increased, and the difference was statistically significant compared with model group(P〈0.05). The NO content in TCM compound group was reduced, and the difference is statistically significant compared with model group(P〈0.01). Conclusion The TCM compound group(Wenrun Tongmai prescription) could increase the SOD activity and NO content, which indicated that it reduced arrhythmia by removing oxygen free radicals,improving the activity of antioxidant enzymes and the ability to resist lipid peroxidation.
