传统中医理论认为陈皮"陈久者良",前期研究从久贮陈皮中分离鉴定了25株真菌,分属于2属5种。研究首次将真菌黑曲霉返接种到无菌陈皮样品中,恒温恒湿培养后,采用HPLC法对陈皮接种黑曲霉培养前后样品芸香柚皮苷、陈皮苷、川陈皮素与桔皮素4种黄酮类成分的变化进行了定量研究;采用紫外-可见分光光度法测定了陈皮接种黑曲霉培养前后样品总黄酮含量变化;采用GC-MS法分析了陈皮接种黑曲霉培养前后样品挥发油类成分的变化。结果发现,陈皮样品接种黑曲霉培养后,橙皮苷等4种黄酮总量及陈皮中所含总黄酮含量均高于接种培养前,挥发油总含量降低,但挥发油成分类型增加,并产生了新的化学成分。研究首次发现黑曲霉代谢转化与陈皮药效物质变化具有相关性,黑曲霉的代谢转化具有促进陈皮药材药效物质积累增加的作用。从真菌黑曲霉与宿主陈皮药效物质基础变化的角度阐明"陈久者良"科学内涵。
We used the fungus Aspergillus niger and inoculated into a sterile sample Citri Reticulatae Pericarpium(PRC) the first time in this paper at the same temperature and humidity condition. The HPLC method was used to determined the contents of four flavonoids. The UV spectrophotometry method was used to determined the total contents of flavonoids. The LC-MS method was used to determined and analysis the contents of volatile oil. We found that the contents of four flavonoids and total flavonoids of PRC were both increasing after the fungus Aspergillus niger was inoculated, and the volatile oil was reducing, but the number of chemicals were increasing of volatile oil. The fungus Aspergillus niger had the effection of accumulate chemical material in PRC. So, "the older, the better" of PRC had its scientific reason, the metabolic activity of Aspergillus niger had a close relation with storage of PRC.
