目的建立汉族人群代谢酶基因分型的永生化淋巴细胞库,为单核苷酸多态(SNP)结构对毒物诱导的基因功能表达的影响建立研究平台。方法用EB病毒(Epstein—Barr Virus,EBV)转化中国汉族健康成人外周血B淋巴细胞,通过对中国汉族人群CYP2E1和NQ01的单核苷酸多态(SNP)位点信息分析,选择中国人群特异性CYP2E1、NQ01基因SNP位点CYP2E1rs2070673,CYP2E1rs2031921,CYP2E13813866,NQO1rs1800566,NQO1rs10517,应用适配器连接介导的等位基因特异性扩增法(ALM-SAS)对上述位点进行基因分型。将供体信息和细胞株基因分型的信息录入,构建中国汉族基因分型的永生化淋巴细胞库。结果通过对细胞株进行上述5个SNP位点的基因分型,获得每株细胞的基因分型信息,获得CYP2E1rs2070673,CYP2E1rs2031921,CYP2E13813866,NQO1rs1800566,NQO1rs10517不同基因分型的永生化淋巴细胞株。结论通过永生化细胞库的建立、基因分型,初步建立中国的汉族人群代谢酶基因分型永生化淋巴细胞库,该细胞库的进一步建设,可以为研究中国人群环境毒物和药物代谢酶基因结构和功能的关系提供技术平台。
Objective To establish immortalized human lymphocytes cell lines with multiple SNP genotyping of Chinese Han population which could afford a platform to study gene function expression influenced by SNPs structure induced by chemical. Methods EBV (Epstein-Barr Virus) were used to immortalize peripheral B lymphocytes derived from Chinese Han population. After analyzing CYP2E1,NQO1 SNP database of Chinese population, the five Chinese specific SNP sites : CYP2E1rs2070673, CYP2E1rs2031921, CYP2E13813866,NQO1rs 1800566,NQOlrs10517 were genotyped by an adapter-ligation mediated allele specific amplification (ALM-ASA) method. The information of providers and genotypes of cell lines were input to establish the database of immortalized human lymphocytes cell lines with multiple SNP genotyping. Results Ninety-six immortalized B lymphoeytes lines were established. The 5 SNPs information of each cell line were gained according to the result of genotyping. Thus, the cell lines with different genotypes on CYP2E1rs2070673, CYP2E1rs2031921, CYP2E13813866,NQO1rs 1800566,NQO1rs10517 SNP sites were obtained. Conclusion An important work has been done in the present paper for the cell bank establishment of the immortalized human lymphocytes cell lines with multiple SNP genotyping based on the establishment of immortalized human lymphocytes cell lines, which may contribute to the research of toxicozenomies and oharmacogenomics.
