目的探讨miR-195对胃癌细胞增殖、迁移及侵袭的影响。方法以人胃癌细胞系SGC-7901细胞株及BGC-823细胞株为研究对象,分别转染miR-195模拟物及抑制物。应用实时荧光定量PCR技术,检测转染后miR-195 mRNA的表达水平。通过CCK8方法检测miR-195对胃癌细胞增殖的影响。通过Transwell实验检测miR-195对胃癌细胞迁移和侵袭的影响。结果 (1)CCK8实验研究显示,与阴性对照(无关序列)相比,转染miR-195模拟物后,SGC-7901细胞的增殖能力减弱;转染miR-195抑制物后,BGC-823细胞的增殖能力明显增强。(2)Transwell小室实验研究显示,与阴性对照相比,转染miR-195模拟物后,SGC-7901细胞的迁移及侵袭能力明显下降;转染miR-195抑制物后,BGC-823细胞的迁移及侵袭能力明显增强。结论 miR-195在胃癌细胞的增殖、迁移及侵袭中发挥重要的作用,miR-195可能成为治疗胃癌的靶基因。
Objective To investigate the effect of microRNA-195( miR-195) on the proliferation,migration and invasion of gastric cancer cells. Methods Human gastric cancer cell lines,including SGC- 7901 cells and BGC-823 cells,were selected,and were transfected with miR- 195 mimics and inhibitors,respectively. The expression level of miR- 195 mRNA was assessed using real time fluorescent quantitative PCR after transfection. Through the CCK8 method and transwell assay,the effect of miR- 195 on migration and invasion of gastric cancer cells was analyzed. Results According to CCK8 test,significant inhibition on the proliferation of SGC- 7901 cells was produced by transfection of miR-195 mimics compared with the negative control group( unrelated sequence); while significant enhancement in proliferation of BGC- 823 cells was observed. According to transwell assay,compared with negative control group,transfection of miR-195 mimics significantly inhibited the migration and invasion of SGC- 7901 cells; while transfection of miR- 195 inhibitor enhanced them. Conclusion miR- 195 play an important role on proliferation,migration and invasion of gastric cancer cells,suggesting a potential targeted gene in therapy of gastric cancer.
