确定了离体法提取时间,并采用离体法对5种海洋微藻硝酸还原酶的特性进行了初步研究。结果表明,离体法的最佳提取时间为5min;在pH7.9及20℃条件下,各藻硝酸还原酶的酶促反应恒速时间及以KNO。为底物的米氏常数存在差异,但以NADH为底物的米氏常数基本相同。酶促反应恒速时间顺序为锥状斯氏藻〈尖刺拟菱形藻〈东海原甲藻〈中肋骨条藻〈旋链角毛藻,以KNO。为底物的米氏常数大小顺序为旋链角毛藻〈东海原甲藻~中肋骨条藻〈锥状斯氏藻~尖刺拟菱形藻。此外,FAD对各藻硝酸还原酶活性的影响存在种属差异。
Nitrate reductase (NR) characteristics of five marine microalgae were investb gated by using in vitro method with determined sample homogenized time. The results showed that the optimum homogenized time was 5 min. On the condition of pH 7.9 and 20 ℃, the constant velocity time of NR catalyzed-reaction and the Michaelis constant (Kin) values for nitrate were different among various species, whereas the Km values for NADH were basically alike. The constant velocity time of NR catalyzed-reaction for different species was in the sequence of Scri p psiella trochoidea 〈 P seudo-nitzsch ia pungens 〈 Prorocentrum donghaiense 〈 Skeletonema costatum℃haetoceroscurvisetus. The order of Km for KN03 for different species was as fol- lows : Chaetoceros curvisetus 〈 Prorocentrum donghaiense 〈 Skeletonema costatum 〈 Scri ppsiel- la trochoidea 〈 Pseudo-nitzschia pungens. In addition, there also appeared species-specific difference in the effects of FAD on NRA.
