目的:观察大鼠牙周改建中白血病抑制因子(leukemiainhibitoryfactor,LIF)与白血病抑制因子受体(leu-kemiainhibitoryfactorreceptor,LIFR)的表达变化。方法选用24只6-7周龄雌性Wistar大鼠,每只大鼠上颌左右两侧分别为实验侧和对照侧,建立大鼠实验性牙移动的模型。将大鼠随机分成3组,分别在加力后3、7、14d处死动物。制备标本,用免疫组化染色法检测LIF和LIFR在大鼠牙周组织中的表达,并采用计算机图像分析软件ImageJ对各组LIF和LIFR在牙周组织中的表达强度进行定量分析。结果LIF和LIFR在正常牙周膜中表达较弱,且主要分布在成纤维细胞、血管内皮细胞、破骨细胞、成骨细胞之中,加力后,实验组表达显著增加。且LIF和LIFR各天之间表达强弱各不相同。在压缩侧,LIF和LIFR的表达在第14天最强,其中LIF第14天的表达显著高于第7天(P<0.05);LIFR在第14天的表达亦显著高于第3天(P<0.01)和第7天(P<0.01)。在牵张侧,LI-FR在第14天表达最强,且比第3天和第7天表达显著增高(均P<0.01),而LIF表达在各天之间无明显的统计学差异。结论LIF和LIFR的表达呈规律性的变化,参与了牙齿移动过程中的牙周组织改建。
Objective To investigate the changes in the expression of leukemia inhibitory factor (LIF)and leukemia inhibitory factor receptor(LIFR)in periodontal remodeling in rats.Methods Experimental model of tooth movement was established in 24 female Wistar rats aged 7 weeks. The left side of upper jaw of each rat was set as experimental side,and the right side as control side.The model rats were randomly divided into three groups,and then were sacrificed after 3,7 and 14 days,respectively.The upper jaw and periodontal tissue were selected for specimen prepa-ration.The expression of LIF and LIFR in periodontal tissue was detected by immunohistochemi-cal staining and was quantitatively analyzed by image analysis software Image J.Results The ex-pression of LIF and LIFR was low in normal periodontal membrane and was mainly distributed in fibroblasts,endothelial cells,osteoclasts and osteoblasts.After force strengthening,the expression of LIF and LIFR significantly increased in experimental group.On the compression side,the ex-pression of LIF and LIFR was highest on the 14th day.Furthermore,the expression of LIF on the 14th day was significantly higher than that on the 7th day and the expression of LIFR on the 14th day was significantly higher than that on the 3rd day and 7th day(P〈0.05 orP〈0.01).On the stretch side,the expression of LIFR was highest on the 14th day.Moreover,the expression of LIFR on the 14th day was significantly higher than that on the 3rd day and 7th day(P 〈0.01).In addition,no significant differences in LIF expression were found among different time points.Conclusion The expression of LIF and LIFR show regular changes in periodontal tissue in rats,suggesting that LIF and LIFR may play im-portant roles in periodontal remodeling during tooth movement.
