母鸡贮精腺位于输卵管的子宫阴道交接部和伞部,为探究鸡碳酸酐酶4基因在输卵管不同部位的表达情况,以探明该基因是否在母鸡贮精腺部位高表达,为进一步研究禽类贮精腺的贮精机理奠定基础。参考鸡CA IV基因序列,在其保守区设计特异性引物,并以鸡β-actin基因为内参基因建立荧光定量方法,基于荧光定量分析CA IV基因在鸡输卵管伞部、膨大部、峡部、子宫部、子宫阴道交接部和阴道部的表达。结果表明,子宫阴道交接部的CA IV基因表达量最高,伞部次之,在膨大部、峡部、子宫部和阴道部表达量较低。CA IV基因在子宫阴道交接部的表达量显著高于在伞部、膨大部、峡部、子宫部和阴道部的表达(P<0.05),在伞部的表达量显著高于膨大部、峡部、子宫部和阴道部的表达(P<0.05)。CA IV基因在母鸡输卵管子宫阴道交接部和伞部位的高表达预示着其参与贮精腺内的精子储存。
The sperm storage tubules of chicken are located at utero-vaginal junction and infundibulum.This study was to explore the expression of CA IV in the different parts of oviduct in chicken,hoping to collect basic data for the mechanism of the avian sperm storage.According to the sequence of chicken CA IV gene,the Real-time PCR primers were designed to amplify the conserved fragment.The β-actin gene was chosen to normalize the expression level of CA IV gene.The established detection method was used to analyze the expression pattern of CA IVgene in the different parts of oviduct in chicken,including infundibulum,magnum,isthmus,uterus,utero-vagina junction and vagina.The results showed that CA IV expression was the highest in the utero-vaginal junction,and its expression was higher than that of the infundibulum(P <0.05).CA IV expression of the utero-vaginal junction and infundibu-lum was higher than that of magnum,isthmus,vagina and uterus(P <0.05).High expression of CA IV in utero-va-ginal junction and infundibulum suggested that CA IV might play an important role in avian sperm storage.
