[目的]以赤子爱胜蚓为原料,研究其体腔液的抗病毒活性及可能的机制,为新型抗病毒药物的开发提供思路。[方法]用分光光度法和琼脂糖凝胶电泳法测定蚯蚓体腔液蛋白酶活性和核酸酶活性;用鸡胚培养检测其抗流感病毒的活性,乙型肝炎病毒表面抗原及e抗原诊断试剂盒检测其对乙型肝炎病毒表面抗原及e抗原的破坏作用。[结果]蚯蚓体腔液具有蛋白酶活性、核酸酶活性和抗病毒活性;各种不同浓度的蚯蚓体腔液对流感病毒的增殖都有显著的抑制作用,且可以抑制乙型肝炎病毒表面抗原和e抗原表达,呈浓度依赖关系。[结论]蚯蚓体腔液具有抗病毒活性,其可能机制是蛋白酶、核酸酶及各种活性成分单独和/或协同作用的结果。
[Objective]To study the antiviral activity of coelomic fluid of eisenia foetida and its possible mechanism,and to provide ideas for the development of new antiviral drugs.[Methods]The protease activity and nuclease activity of coelomic fluid in earthworm were detected by using spectrophotometric method and agarose gel electrophoresis(AGE);chicken embryos cultivation was used to evaluate its anti-influenza viral activity,HBsAg and HBeAg diagnostic kit was used to detect its katogene to HbsAg and HBeAg.[Results]The coelomic fluid of eisenia foetida had protease activity,nuclease activity and antiviral activity;all different concentrations had significantly inhibition to the generation of influenza virus,while the coelomic fluid could inhibit the expression of HbsAg and HBeAg,which revealed a dosage-effect relationship.[Conclusion]The coelomic fluid has antiviral activity,its mechanism may be the solo-action or synergistic effects of protease,nuclease and all sorts of activity ingredients.
