目的:探讨生长抑制因子4(ING4)基因在神经胶质瘤发生发展过程中的作用。方法:采用RT-PCR方法扩增34例神经胶质瘤患者手术标本和24例正常脑组织标本的ING4基因和内参照GAPDH基因.凝胶分析软件对RT-PCR产物进行半定量分析。比较ING4基因mRNA在神经胶质瘤和正常脑组织中的表达情况。结果:所有的标本均扩增出ING4基因的目的条带。神经胶质瘤中的ING4基因mRNA表达水平明显低于正常脑组织(P〈0.05)。恶性度高的神经胶质瘤组明显低于低度恶性神经胶质瘤组(P〈0.05)。结论:ING4基因在神经胶质瘤的发生发展过程中可能起一定的作用.且与恶性程度有一定相关性。
Objective To investigate the role of ING4 gene in the tumorigenesis and development of glioma.Methods The mRNA expressions of ING4 gene and intra-reference GAPDH gene in 34 specimens of glioma tissue and 24 of normal brain tissue were assessed by reverse transcription polymerase chain reaction (RT-PCR), Results The target bands of ING4 gene were present in all of the specimens. The mRNA expression level of ING4 gene was significantly lower in glioma than in the normal brain tissue (P 〈 0.05), and the level in the high-grade malignancy was markedly lower than that in the low-grade malignancy (P 〈 0.05). Conclusion ING4 gene may play a role in the tumorigenesis and development of glioma and be related to the grade of the malignancy.
