目的:探讨兰索拉唑对幽门螺杆菌(helicobacter pylori,Hp)感染的胃上皮细胞的体外抗炎效应。方法体外培养胃上皮细胞系AGS,加入兰索拉唑和培养的Hp感染不同时间。ELISA检测IL-1β和IL-8的产生,Western blot检测NF-κB p 65亚基的核转位以及红素氧合酶1(Heme oxygenase-1, HO-1)蛋白表达水平。结果兰索拉唑孵育8 h后,能显著抑制Hp诱导的IL-1β和IL-8。同时,兰索拉唑能抑制NF-κB p 65亚基的核转位,并能上调HO-1蛋白的表达。结论兰索拉唑可能通过抑制NF-κB的激活,上调HO-1的表达而发挥对胃上皮细胞的抗炎作用。
Objective To investigated the anti-inflammatory effects of Lansoprazole in H. pylori-infected gastric epithelial cells in vitro. Method Gastric epithelial cell line AGS was cultured in vitro, and was incubated with Lansoprazole and H. pylori for various times. Production of IL-6 and IL-8 were detected by ELISA, activation of NF-κB and expression of Heme oxygenase-1 was detected by Western blot. Results After incubated for 8 h, Lansoprazole could significantly inhibit H. pylori induced IL-1βand IL-8 production. And Lansoprazole could also induce AGS cells expression of HO-1. Furthermore, translocation of NF-κB p 65 subunit was inhibited, as compared with H. pylori group. Conclusion Lansoprazole could inhibit activation of NF-κB and induction of HO-1, and thus account for the gastroprotective effects in vasoprotection.
