【目的】建立在大鼠血浆中测定毛冬青酸血药浓度的方法。【方法】大鼠尾静脉注射毛冬青酸后,依一定的时间间隔,自颈动脉采血,分离血浆,经氯仿沉淀蛋白后,以乙腈一体积分数0.2%磷酸为流动相,色谱柱为Kromasil 100—5 C18(250mm×4.6mm,5μm),流速为1.0mL/min;柱温为30℃;紫外检测波长为203nm。【结果】本研究方法在2.04~51.0μg/mL范围内线性关系良好,低、中、高3个浓度的回收率为93.5%~94.8%,日内精密度sR为1.5%~4.5%、日间精密度SR为2.7%~11.1%。【结论】经过系统的方法学考察,本研究所建立的测定方法灵敏度较高、重现性良好,可用于血浆中毛冬青酸的含量测定。
Objective To establish a method for the determination of ilexolic acid in rat plasma. Methods Blood sample was taken timely from the carotid artery of rats receiving injection of ilexolic acid through caudal vein. Plasma was isolated and the protein was precipitated by chloroform. Then the plasma concentration of ilexolic acid was determined with ultraviolet-HPLC. The chromatographic conditions were as follows: Kromasil 100-5 C18 column (250mm ×4. 6mm, 5μm) at 30℃, a mixture of acetonitrile - 0.2% phosphoric acid (V/V) as the mobile phase, in gradient elution, at the flow rate of 1.0 mL/min and UV detection wavelength being 203 nm. Results The linear range of ilexolic acid was 2.04 -51.0 μg · mL^-1 , the average recoveries of ilexolic acid at low-, moderate- and high-concentrations were 93.5% -94. 8%, and the intra-day RSD was 1.5% -4.5% and inter-day RSD was 2. 7% -11.1%. Conclusion The method has been confirmed to be simple, stable, reproducible and with high specificity, and can be used for the pharmacokinetic study of ilexolic acid in rats.
