钙粘蛋白(cadherin)是许多鳞翅目昆虫中苏云金芽孢杆菌(Bt)毒素的受体。利用RNAi技术对小菜蛾中肠cadherin基因进行沉默处理,明确了cadherin基因沉默对小菜蛾对Cry1Ac毒素敏感性的影响。结果表明,注射目标dsRNA后的第1天至第5天,Cry1Ac敏感及抗性小菜蛾品系cadherin的基因表达量均显著减少,尤以第2天的减少量最为明显;Cry1Ac毒力测定结果表明,沉默cadherin基因能在一定程度上提高幼虫的成活率,即可使小菜蛾对Cry1Ac毒素的敏感性降低。研究初步表明,cadherin基因的功能与小菜蛾对Cry1Ac毒素的抗性有关。
Cadherin was confirmed to be the receptor of Bacillus thuringiensis (Bt) insecticidal protein in several Lepidoptera insects. Cadherin gene in Plutella xylostella was silenced by RNAi technique and effect of cadherin gene silencing on susceptibility of P. xylosteUa larvae to CrylAc toxin was investigated. The results showed that the expression level of cadherin gene reduced from the first day to the fifth day after the dsRNA corresponding to cadherin segment was injected, and the most significant reduction was observed sharply at the second day. The susceptibility of resistant and susceptible strain of P. xylostella larvae to CrylAc toxin decreased after cadherin gene being silenced. The results suggested that cadherin gene was related to resistance of P. xylostella to CrylAc toxin.
