目的:研究青蒿琥酯(ART)对增生性瘢痕来源的成纤维细胞(HFBs)的Ⅲ型胶原(COLⅢ)蛋白表达的影响。方法:原代培养人HFBs,分为空白对照组和ART干预组(分别用75μmol/L、150μmol/L、300μmol/L、600μmol/L ART干预24h),荧光定量PCR(q-PCR)法检测Ⅲ型前胶原α1链(proCOL3A1)的基因转录情况,western blotting法检测COLⅢ蛋白的表达,酶联免疫吸附试验(ELISA)检测基质金属蛋白酶1(MMP-1)的含量。结果:与空白对照组比较,ART干预组的proCOL3A1 mRNA相对表达量明显降低,且不同浓度ART组间比较差异有统计学意义(P〈0.01)。ART干预组COLⅢ蛋白表达水平明显下降,细胞培养上清液中MMP-1含量明显升高,且各浓度组间两两比较差异均有统计学意义(均P〈0.05或P〈0.01)。结论:ART能抑制人HFBs的COLⅢ的基因转录和蛋白水平,同时促进MMP-1分泌增加,加速胶原降解,抑制细胞外基质沉积,这可能是ART治疗增生性瘢痕的机制之一。
Objective: To investigate the effect of artesunate (ART) on the expression of type Ⅲ collagen (COL Ⅲ) in hypertrophic scar fibroblasts (HFBs). Methods: HFBs isolated from tissue specimens of hypertrophic scar were treated with different concentrations (75, 150, 300 and 600 μmol/L) of ART for 24 h. The mRNA expression of proCOL3A1 was detected by fluorescent quantitative PCR (q-PCR). The protein expression of COL Ⅲ was detected by western blotting. The matrix metalloproteinase (MMP) 1 level in supernatants was assessed by enzyme-linked immuno-sorbent assay (ELISA). Results: The pro- COL3A1 mRNA level and the COL Ⅲ protein expression level were decreased, while the MMP-1 level in supernatants of culture medium was increased in ART groups as compared with the control group ( P 〈 0.01). There were statistical differences in above indexes among different ART concentration groups ( P 〈0.05 or P 〈0.01). Conclusion: ART may have a curative effect on hypertrophic scar, and the mechanisms may be related to the inhibition of MMP-1 release and the regulation of COL Ⅲ expression.
