【目的】通过对缺血再灌注早期脑组织的内皮型一氧化氮合酶(endothelial nitric oxide synthase,eNOS)表达情况的观察,探讨一氧化氯在脑缺血再灌注损伤早期发挥神经毒性作用时eNOS亚型的变化。【方法】采用线栓法制作大鼠脑缺血再灌注模型,免疫纽化及western blot方法检测缺血脑组织的eNOS蛋白表达变化。【结果】免疫组化及western blot结果显示缺血区脑组织血管内皮细胞内的eNOS表达在缺血1h内达高峰,缺血2h到再灌注2h内持续降低。【结论】大鼠脑缺血再灌注模型中血管内皮细胞eNOS出现变化,表明一氧化氮在缺血性脑损伤的病理过程的作用发挥与eNOS亚型的变化有关。
[Objective] To explore the change of the subtypes of endothelial nitric oxide synthase(eNOS) while nitric oxide(NO) plays neurotoxic effect in the early stage of cerebral ischemia-reperfusion through observing the expression of eNOS in brain tissues of rats during cerebral ischernia-reperfusion. [Methods] Cerebral ischemia-reperfusion injury was induced by 2-hour suture occlusion of the unilateral middle cerebral artery, immediately after suture withdrawal to allow reperfusion. The eNOS expressions were examined by the method of immunohistochemistry and western blot. [Results] The eNOS expression increased in 1-hour during ischemia, keeping up with decreasing until reperfusion 2-hour. [Conclusion]The expression of eNOS in rats changes during cerebral ischemia-reperfusion. It indicates that the role of NO in pathogenesis of cerebral ischemic injury may be related to the change of eNOS subtypes.
