目的研究肥大细胞类胰蛋白酶(tryptase)对小鼠脑微血管内皮细胞系bEnd-3表达白细胞介素-6(IL-6)和肿瘤坏死因子-α(TNF-α)的影响以及可能的信号途径。方法bEnd-3细胞与不同浓度的类胰蛋白酶共同培养后,用RT-PCR和放射免疫的方法检测细胞及其上清液中IL-6、TNF-α mRNA和蛋白的表达量以及蛋白酶激活受体2(PAR-2)对其表达的影响。结果类胰蛋白酶上调bEnd-3 IL-6和TNF-α mRNA的表达和分泌,并呈时间和剂量依赖性,这一作用可能是通过激活细胞膜上的PAR-2而发挥的。结论肥大细胞类胰蛋白酶参与bend-3微血管内皮细胞的致炎性反应。
Purpose To study the effect of mast cell tryptase on the expression of IL-6 and TNF-α in bend. 3 (a mouse brain microvascular endothelial cell strain). Methods bend. 3 cell strains were cultured with different concentrated tryptase. After isolating total RNA, reverse transcription polymerase chain reaction (RT-PCR) was made to examine the mRNA level of IL-6 and TNF-α in cells. Meanwhile,tryptase-induced cytokine secretion in the cultured supernatants was measured by radioimmunoassay. The effect of PAR-2 on the expression of IL-6 and TNF-α in bend. 3 was also explored. Results Tryptase induced up-regulation of the expression of IL-6, TNF-α mRNA and protein in concentration-dependent and time-dependent manners by activating protease-activated receptor 2 (PAR-2). Conclusions Mast cell tryptase may be involved in pro-inflammatory reaction in microvascular endothelial cell.
