目的:探讨脂肪来源间充质干细胞( ADSCs)移植在全层皮肤缺损修复中的作用。方法25只SD大鼠制作大鼠背部全层皮肤缺损模型,随机分为3组,分别移植胶原膜(胶原膜组)、ADSCs+胶原膜( ADSCs+胶原膜组)后覆盖异体SD大鼠皮肤及直接以异体SD大鼠皮肤覆盖(空白对照组)。观察各组创口的愈合速度,并通过愈合皮肤抗拉力实验、HE染色、原位杂交的方法检测愈合后皮肤的抗拉性、组织学改变及ADSCs的表达。结果 ADSCs及胶原膜对大鼠一般情况无明显影响;ADSCs+胶原膜组创面愈合率为(94.24±3.13)%,显著高于空白对照组的(84.57±2.42)%及胶原膜组的(84.72±9.70)%,差异有统计学意义(P<0.05);ADSCs+胶原膜组皮肤的最大拉力、弹性位移和拉伸率与空白对照组及胶原膜组比较差异无统计学意义( P>0.05);ADSCs+胶原膜组皮肤真皮层中存在大量类似于正常皮肤的毛囊结构;在ADSCs+胶原膜组愈合创面皮肤组织中可追踪到雄性大鼠来源细胞的阳性表达。结论 ADSCs能促进大鼠全层皮肤缺损的修复。
Objective To investigate the effects of adipose -derived stem cells ( ADSCs ) transplantation in full thickness cutaneous deficiency repairing in rats .Methods Full thickness cuntaneous deficiency models were constructed in 25 SD rats and randomly divided into control group , collagen membrane group ( negative control ) and ADSCs-collagen membrane group;in which allogeneic skin graft ( ASG) alone, ASG plus collagen membrane , and ASG plus ADSCs-col-lagen membrane were given for wound covering , respectively.The level of wound healing was observed .Tensile test, HE staining and in situ hybridization were carried out to assess the tensile strength , histological changes and ADSCs′expres-sion in new skin , respectively .Results Animal experiment showed that ADSCs and collagen membrane transplantations were safe.The wound healing rate of ADSCs -collagen membrane group [(94.24 ±3.13)%] was significantly higher than those of control group [(84.57 ±2.42)%] and collagen membrane group [(84.72 ±9.70)%].There was no sig-nificant difference was revealed in the tensile strength of new skin among the three groups ( P〉0.05 ) .HE staining showed that there were neogenetic skin appendages in experimental group rather than controls .According to in situ hybrid-ization, SRY DNA was positive in the new skin of ADSCs -collagen membrane group .Conclusion ADSCs transplanta-tion can promote the full thickness cutaneous deficiency repairing in rats .
