目的 研究香兰素吸嗅对大鼠抑郁样行为改善及其机制。方法 慢性不可预见性中等强度应激(CUMS)+孤养的方法建立大鼠抑郁症模型。为验证香兰素作用途径,另设立大鼠嗅球毁损(OBX)模型。于造模前、后及干预后4 w测试大鼠糖水消耗量和强迫游泳不动时间,干预结束后超高压液相色谱检测大鼠脑内5-HT、DA、NE的量。结果 与造模后相比,CUMS+香兰素吸嗅组大鼠神经行为学指标显著好转(P〈0.05);其脑匀浆液5-HT、DA的量显著高于空白对照组(P〈0.05)。与造模后相比OBX+香兰素吸嗅组大鼠神经行为学指标未见好转(P〉0.05);其脑匀浆液5-HT、DA、NE均显著低于假手术组(P〈0.05)。结论 香兰素可经嗅觉通路改善大鼠抑郁样行为,其机制可能通过提升脑内5-HT、DA实现的。
Objective To explore the effects of vanillin sniffing to behavioral of depressive model rats and the possi- ble mechanism. Methods CUMS + solitary ways were used to support the establishment of depression model. In order to identify the pathway by which vanillin worked ,olfactory bulbeetomy + vanillin group(OBX) were added. Forced swimming test(FST) and Sucrose consumption test(SCT) were conducted before and after CUMS( or operation) ,and at the end of drug administration. Then,the level of brain 5-HT, DA, NE were detected by Ultra Performance Liquid Chromatography (UP- LC). Results Compared with post-CUMS, the neurobehavioral index improved significantly in CUMS + vanillin sniffing group( P 〈 0.05 ) ;and the level of brain 5-HT, DA were significantly higher than the control group (P 〈 0.05 ). Compared with post-CUMS, the neurobehavioral index was no significant changes in OBX + vanillin sniffing group (P 〉 0.05 ) , and the level of brain 5-HT, DA, NE were all significantly lower than the sham group( P 〈 0.05 ). Conclusion Vanillin may re- lieve the depression-like behavior in rats by olfactory pathway;The mechanism may be increasing the level of brain 5-HT and DA.
