目的研究片段睡眠(SF)对小鼠垂体、下丘脑和枕叶皮质的精氨酸加压素(AVP)和神经肽Y(NPY)含量的影响及莫达非尼对SF模型小鼠上述指标的影响。方法80只雄性昆明种小鼠随机分为对照(C)组、SF组、莫达非尼(M)组和SF+莫达非尼(S+M)组。应用SF剥夺平台.制作SF小鼠模型,SF组小鼠和S+M组小鼠进行72h的SF。M组小鼠和S+M组小鼠灌胃给予莫达非尼420mg·kg^-1·d^-1×3d。应用放射免疫分析法测定小鼠垂体、下丘脑和枕叶皮质的AVP和NPY含量。结果SF模型小鼠垂体、下丘脑和皮质AVP和NPY的含量比C组小鼠显著减少(P〈0.01)。经过莫达非尼干预的SF模型小鼠,垂体、下丘脑和枕叶皮质AVP和NPY的含量也比C组小鼠显著减少(P〈0.05或P〈0.01).但与SF模型小鼠比较无显著差异(P〉0.05)。结论SF减少小鼠垂体、下丘脑和枕叶AVP和NPY的含量,而莫达非尼干预对SF模型小鼠脑内AVP和NPY的含量无显著影响。
AIM To investigate the effects of sleep fragmentations (SF) on arginine vasopressin (AVP) and neuropeptide Y (NPY) contents in the tissues of pituitary, hypothalamus and occipital cortex in mice and the changes of AVP and NPY contents in these tissues in modafinil-treated SF model mice. METHODS Eighty male Kunming mice were randomly divided into control (C), SF model, modafinil (M), and SF + modafinil (S + M) groups. A mouse model of SF was prepared by means of sleep deprivation experimental equipment. Seventy-two hours SF was conducted in both SF and S + M mice. Modafinil (420 mg'kg-l"d-1) was administered intragastrically once daily for 3 days in both M and S + M mice. Radioimmunoassay was used to analyze the contents of AVP and NP~ in the tissues of pituitary, hypothalamus and occipital cortex in mice. RESULTS The contents of AVP and NPY were significantly decreased in the tissues of pituitary, hypothalamus and occipital cortex of both SF model (P 〈 0.01) and S + M mice (P 〈 0.05, P 〈 0.01), compared with those of control mice. There were no significant differences in the contents of AVP and NPY in pituitary, hypothalamus and occipital cortex between SF model mice and modafinil- treated SF mice (P 〉 0.05) . CONCLUSSION The contents of AVP and NPY in pituitary, hypothalamus and occipital cortex of mice is decreased by SF, which is not significantly changed by the treatment with modafinil.
