在碱性介质中,6-巯基嘌呤(6-MP)与过氧化氢反应可在405nm波长处产生微弱的荧光,微量铜(Ⅱ)的存在能大大增敏其荧光强度。据此,建立了一种高灵敏测定6-MP的荧光光度分析新方法。在优化的条件下,体系的荧光强度与6-MP的浓度在2.1×10^-8~1.7×10^-5mol·L^-1范围内呈线性关系,检出限为7.1×10^-9mol·L^-1。方法用于片剂中6-MP含量的测定,其结果与药典方法对照,令人满意。
In basic media, 6-mercaptopurine reacts with hydrogen peroxide to form a new fluorescent compound,which possesses a weak emission wavelength of 405 nm. Its fluorescence intensity can be enhanced remarkably when trace Cu^2+ exists as catalyst. Based on this, a novel spectrofluorimetric method with high sensitivity was developed for the determination of 6-mercaptopurine. Under the optimal conditions,the calibration graphs are linear over the range of 2.1 ×10^-8~1.7 ×10^-5 mol·L^-1 with a detection limit of 7.1×10^-9 mol·L^-1. The proposed method was successfully applied to determine 6-mercaptopurine in pharmaceutical preparations, the results obtained by pharmacopoeia method.
