目的观测CXCR4活化对人恶性胶质瘤细胞系U87中血管内皮生长因子(VEGF)、白细胞介素-8(IL-8)基因表达及蛋白分泌的影响。方法采用间接免疫荧光标记观测CXCR4在U87细胞中的表达和定位;用间质细胞衍生因子-1α(SDF-1α)激活CXCR4,通过酶联免疫吸附试验(ELISA)检测U87细胞培养上清中VEGF、IL-8蛋白的含量,用逆转录聚合酶链反应(RT-PCR)检测二者mRNA的表达。结果CXCR4表达于U87细胞的胞膜和胞质,经活化后可增加VEGF、IL-8mRNA的表达和蛋白分泌量。结论人恶性胶质瘤细胞中CXCR4活化后能够促进血管生成因子VEGF和IL-8的产生。
To investigate the effects of activating CXCR4 by its ligand, stromal cell-derived factor-lot in human glioblastoma U87 cells on the changes of vascular endothelial growth factor and interleukin-8. Methods Expression and localization of CXCR4 in U87 cells were detected by indirect immunofluorescence staining. The production of vascular endothelial growth factor and interleukin-8 was measured by enzyme-linked immunosorbent assay and the mRNA was detected by reverse transcription polymerase chain reaction in U87 cells with or without stimulation of stromal cell-derived factor-1α. Results CXCR4 was mainly expressed on the plasma membrane and cytoplasm of U87 cells. Activation of CXCR4 by SDF-1α increased IL-8 and VEGF protein secretion and mRNA expression. Conclusion Activation of CXCR4 promotes malignant glioma cells producing vascular endothelial growth factor and interleukin-8, which might contribute to the angingenesis of the tumor.
