直观的观察雷氏大疣蛛毒素促进神经胶质瘤U87凋亡的形态学变化。首先用蛋白印迹法检测合适的雷氏大疣蛛毒素浓度处理U87细胞,再用该浓度毒素处理U87细胞不同时间;用100pg/m L的雷氏大疣蛛毒素作用48 h诱导神经胶质瘤U87细胞凋亡,然后用倒置相差显微镜,Gimsa染色法,吖啶橙/嗅化乙锭(AO/EB)双重荧光染色法以及流式细胞法,从最直观的形态学上观察雷氏大疣蛛毒素促进神经胶质瘤U87凋亡的形态学变化。实验组的细胞出现很明显的凋亡小体,细胞膜不会破裂,细胞质不会流出,细胞核含有固缩核片段等细胞凋亡的形态学上的特征。雷氏大疣蛛毒素作用神经胶质瘤U87细胞后的形态学的变化,基本包括了细胞凋亡的主要的形态学特征。说明雷氏大疣蛛毒素是通过促进神经胶质瘤U87细胞凋亡从而抑制细胞的增殖。
The morphological changes of U87 glioma cells apoptosis promoted by Macrothele raveni toxin was intuitive observation. Firstly,a suitable toxin concentration was detected by western blot,and then U87 glioma cells were conducted by the concentration with different processing times. 100 pg / m L of toxin for 48 h was used to induce U87 glioma cells apoptosis. The morphological changes were intuitive observation with inverted phase contrast microscope,Gimsa staining,acridine orange / ethidium bromide( AO / EB) double fluorescent staining and flow cytometry. The cells in experiment group appeared obvious apoptosis body,and the cell membrane was not break and cytoplasm was not flow out. The morphology characteristics of cell apoptosis appeared on cell nucleus such as Pyknotic nucleus fragments. U87 glioma cells morphological changes treated with Macrothele raveni toxin was basically include the main morphological characteristics of apoptosis. The result proved that Macrothele raveni toxin was inhibit of U87 glioma cells proliferation by apoptosis.
