目的构建pEGFP—Ngb荧光表达载体并研究其在缺氧条件下SY5Y细胞中的功能。方法小鼠NgbeDNA被亚克隆进pEGFP—N1载体,重组载体转染SY5Y细胞,通过双苯并咪唑染料(Hoechest)和碘化丙啶(PI)染色观测细胞在低氧条件的抗缺氧作用。结果成功构建pEGFP—Ngb荧光表达载体,表达Ngb的SY5Y细胞在低氧条件下的存活率增加。结论Ngb在低氧条件下对SY5Y细胞存活可能具有重要作用。
Objective: To construct pEGFP- Ngb expression vector and to investigate its function in SY5Y cells under hypoxia condition. Methods The Ngb eDNA of mice was subcloned into pEGFP - N1 vector and the recombinant vector:was transfected into SY5Y cells to observe its antihypoxic function by Hoechest and PI dyeing. Results: The pEGFP- Ngb, oxpression vector was successfully constructed. The survival rate of SY5Y cells which expressed Ngb was increased under hypoxia condition. Conclusion: Ngb may play an important role in cell survive under hypoxla condition.
