目的 通过了解细胞外信号调节蛋白激酶1 (ERK-1)在缺氧脑损伤大鼠脑中的表达,了解其参与缺氧脑损伤的过程.方法 48只3d龄新生SD大鼠,随机分成2组:对照组和缺氧组,每组各24只,缺氧组SD大鼠置于自制密闭容器中,充以含8%氧气的氧氮混合气体,时间为90 min,对照组SD大鼠不进行处理.分别于造模后6个时间点——1、3、7、14、21 d和28 d后留取两组SD大鼠脑组织,采用Western blot检测ERK-1总蛋白在脑组织中的表达情况.结果 缺氧组大鼠脑组织ERK-1呈现动态表达;变化规律:在缺氧早期(缺氧后1~7 d),ERK-1总蛋白的表达均有降低的趋势,缺氧后第14天开始其表达具有升高的趋势,缺氧后第21天和第28天ERK-1表达量又有所下降,差异无统计学意义(P>0.05).结论 缺氧造成新生大鼠脑损伤时,ERK-1总蛋白的表达谱没有明显变化的趋势,提示在早产儿缺氧性脑损伤时ERK-1并不通过剂量的变化对缺氧后脑组织损伤进行调节.
Objective To understand the process of extracellular signal regulated protein kinase 1(ERK-1) involved in hypoxic brain injury through the understanding of its expression in hypoxic brain injury in rat brain. Methods 48 3-day SD rats were randomly divided into two groups: control group and hypoxia group, 24 rats in each group. Rats of hypoxia group were treated with mixed gas which included O2(8%) and N2 for 90 minutes, and rat pups of the control group were treated with no treatment. Brain tissues of both groups were obtained at the 1st, 3rd, 7th, 14 th, 21 st day and the28 th day after hypoxia insult. Western blot was used to detect the expression of ERK-1 in brain tissues. Results There were dynamic changes of ERK-1 in brain tissue of rats after hypoxia. It showed decreased expression of total protein of ERK-1 at 1-7 days after hypoxia, and on the 14 th day of hypoxia, there seemed to exist a rising trend in the expression of ERK-1. And the expression of ERK-1 decreased again on the 21 st and 28 th day, with no significant difference(P 0.05). Conclusion When hypoxia happened, there is no significant change in the expression profile of ERK-1 protein, suggesting that ERK-1 may not affect brain tissue by dose regulation after hypoxia insult.
