目的 探讨丁酸钠抑制人卵巢癌3AO细胞侵袭转移能力的机制.方法 采用光镜和免疫组织化学染色、流式细胞术观察药物作用后细胞结构及转移抑制基因KAI1表达的变化,逆转录聚合酶链反应实验观察KAI1mRNA表达变化.结果 药物作用后KAI1mRNA表达量显著高于对照组(P〈0.01);KAI1基因蛋白表达较对照组明显增强(P〈0.01),流式细胞术检测KAI1阳性表达率升高(P〈0.05).结论 转移抑制基因KAI1的表达变化可能是丁酸钠抑制人卵巢癌细胞增殖和侵袭的机制之一.
Objective To evaluate the effects of sodium butyrate (NaB) on the metastasis of human ovarian cystadenocarcinoma cell line 3AO. Methods The morphologic changes were observed by reverse phase-contrast microscope, variation of KAI1 gene expression was estimated by immunohistochemistry, reverse-transcription polymerase chain reaction (RT-PCR) and flow eytometrY. Results The expression of KAI1 in NaB group was significantly higher than that in control group by immunohistochemistry and flow cytometry. The result of RT-PCR showed that the expression of KAIlmRNA was higher in NaB group than that in control group. Conclusion The change of KAI1 gene may be a possible mechanism that NaB can inhibit the proliferation and invasion of human ovarian cancer cell line.
