目的本研究对姜黄挥发油抑制皮肤鳞状细胞癌A431细胞增殖的效果进行研究,并分析Notch通路的参与作用,为临床治疗和新药研发提供参考。方法将生长状态良好的皮肤鳞状细胞癌A431细胞随机分为4组:对照组、姜黄挥发油低剂量组(1mmol/L)、中剂量组(10mmol/L)和高剂量组(100mmol/L)(n=8)。除对照组细胞外,其他各组细胞均采用对应剂量的姜黄挥发油共培养4h,分析各组细胞的增殖抑制率,采用酶联免疫吸附测定法检测Notch通路蛋白的表达变化。结果低剂量组、中剂量组和高剂量组的细胞增殖抑制率分别为15.1%、22.9%和39.9%(P〈0.05),同时,随着姜黄挥发油剂量的增加,Bax蛋白的表达明显升高(P〈0.05),而Bcl2蛋白的表达明显降低(P〈0.05),Notch1和Notch2蛋白的表达也明显降低(P〈0.05)。结论姜黄挥发油能有效抑制皮肤鳞状细胞癌A431细胞增殖,且可能与其抑制Notch通路蛋白的表达有关。
Objective The present research aimed to explore the inhibition of turmeric volatile oil on squamous cell carcinoma cell line A431and the involvement of Notch pathway. Methods Squamous cell carcinoma cell lines A431 were divided into 4 groups, control group, turmeric volatile oil at low ( 1mmol/L), middle (10mmol/L) and high dose (100mmol/L) groups (n= 8). The inhibition effect on cell proliferation was detected by MTT and the expression of Notch1 and Notch2 were assayed by real time PCR, ELISA and immunohistochemistry. Results The inhibition rates were 15.1%, 22.9% and 39.9% at low, middle and high doses, showing significant differences (P〈0.05). Along with the increase of the dose of urmeric volatile oil, the expression of Bax was significantly increased while the expression of Bcl2, Notchl and Notch2 was significantly decreased (P〈0.05). Conclusions Turmeric volatile oil inhibits the proliferation of squamous cell carcinoma cell line A431, which may involve inhibition of the Notch pathway.
