目的:初步探究他汀类药物对人结肠癌和肺癌细胞株的毒性作用。方法:以四甲基偶氮唑蓝(MTT)比色法测定辛伐他汀(Simvastatin,SV)和阿托伐他汀(Atorvastatin,AV)对结肠癌细胞株HCT116、肺癌细胞株A549的细胞毒作用,计算两种药物对不同肿瘤细胞的半数抑制浓度(Half maximal inhibitory concentration,IC50)。结果:SV、AV对HCT116的IC50分别为42.992μmol·L-1、47.845μmol·L-1,SV、AV对A549的IC50分别为41.50μmol·L-1、50.06μmol·L-1。结论:SV、AV对两种肿瘤细胞均有抑制作用,抑制率大小呈剂量依赖性,不同他汀类药物对肿瘤细胞的抑制作用存在差异。
Objective:To measure the cytotoxic effects of the statins on human cancer cells.Methods:The cytotoxic effects of the statins on human colon cancer cells HCT116 and lung cancer cells A549 were measured by MTT to determine IC50 values of atorvastatin and simvastatin on HCT116 and A549.Results:IC50values of atorvastatin on HCT116 is 42.992μmol·L-1,IC50 values of simvastatin on HCT116 is 47.845μmol·L-1· IC50 values of simvastatin on A549 is 41.5μmol·L-1,IC50 values of atorvastatin on A549 is 50.06μmol·L-1.Conclusion:SV and AV could inhibit the proliferation of cancer cells.Statins inhibit cancer cells in a dose-dependent manner,different statins have different effects on cancer cells.
