Adipose triglyceride lipase (ATGL) is recognized as a novel triglyceride lipase primarily to catalyze the first step of triglycerlde hydrolysis and plays an important role in energy homeostasis. This review summarizes the current understandings on the structure of the lipase, the physiological functions of ATGL as a hormone-sensitive lipase (HSL), the regulation of ATGL in associated diseases, and finally the remained questions to be investigated.
