目的探讨生肌玉红胶原促进慢性缺血性创面愈合的可能作用机制。方法 72只大鼠随机分为空白组、对照组、实验组3组,每组24只。所有大鼠建立大鼠后肢缺血性创面模型,空白组直接缝合皮肤,实验组以生肌玉红提取液(0.5 g/ml,50μl/只)载入10 mm×10 mm×2 mm长方体胶原放置血管离断缺损处的缺血肌肉组织处,对照组将载入50μl生理盐水的相同大小胶原放置于血管离断缺损处的缺血肌肉组织处,然后缝合皮肤。各组均观察28天。观察各组大鼠一般情况,并分别于第3、7、14、28天检测大鼠后肢缺血创面处微血管计数及对照组与实验组植入标本内血红蛋白含量。结果空白组大鼠后肢均有跛行,部分苍白并见到肢端青紫,实验组大鼠后肢无明显缺血表现。与空白组同时间点比较,对照组第14、28天时微血管计数高于空白组,实验组第7、14、28天时亦明显高于空白组(P〈0.05)。与对照组同时间点比较,实验组第7、14天时微血管计数明显升高(P〈0.05)。实验组第3、7、14、28天植入标本内血红蛋白含量明显高于同时间点对照组(P〈0.05)。结论生肌玉红胶原可促进缺血性创面组织血管新生并改善其血流灌注,可能为其促进慢性缺血性创面愈合的作用机制。
Objective To investigate the possible mechanism of the collagen loading with Shengji Yuhong Onit-ment in promoting chronic ischemic wound healing. Methods Randomly divided 72 rats into the blank group,the control group and the experimental group,with 24 rats in each group. All rats were established into hind limb ischemic wound model.,with direct skin suture in the blank group,the oblong collagen( size: 10 mm 10 mm × 2 mm)was loaded with the Shengji Yuhong extract( 0. 5 g / ml,50 μl / each) in the experimental group and placed it at the ischemic muscle tissue in the blood vessel defect,with the same size collagen loaded with 50 μl normal saline in the ischemic muscle tissue in the blood vessel defect in the control group. Then the skin was sutured. Twenty-eight days were observed in each group. Observed the general situation of rats in each group,detected the microvessel count in the rat' s ischemic wound hind limb and hemoglobin content in implanted samples in the control group and the experimental group on the 3 th,7 th,14 th day respectively. Results The rats in the blank group were hind limb claudication,partly pale and can see acrocyanosis,while rats in the experimental group had no obvious ischemic manifestations. Compared with the blank group,the microvessel count in the control group was higher than the blank group on the 14 th and 28 th day,and the microvessel count in the experimental group was significantly higher than that of the blank group on the 7 th,14 th and 28 th day( P〈0. 05). Compared with the control group,the microvessel count was significantly increased in the experimental group on the 7 th and 14 th day( P〈0. 05). The hemoglobin level of the implanted samples in the experimental group was significantly higher than that of the control group at the same time on the 3 rd,7 th,14 thand 28 th day( P〈0. 05). Conclusion The collagen loading with Shengji Yuhong ointment can promote angiogenesis in ischemic wound tissue and improve blood flow perfusion,which may the
