目的探讨踝蛋白(talin)mRNA在大肠癌组织中的表达变化及其在大肠癌发生、发展中的作用。方法采用荧光定量逆转录PCR(FQ-RT-PCR)方法检测41例大肠癌及癌旁正常组织中talin mRNA的表达。结果talin mRNA在大肠癌组织中的表达量为126535copy/μg RNA,而在癌旁正常组织中为22689376copy/μg RNA,两者相比,P〈0.01;有淋巴结转移的大肠癌较无转移者talin mRNA表达低,P〈0.01;癌组织浸润越深,talin mRNA表达越低,P〈0.01。结论talin mRNA在大肠癌中表达降低;talin与大肠癌的浸润、转移有关,可作为大肠癌发生、发展的预测指标之一。
Objective To investigate the mRNA expression of talin in colorectal carcinoma and the roles of talin in tumorigenesis and progression of colorectal carcinoma. Method The expression of talin mRNA in 41 samples of colorectal carcinoma and adjacent normal tissues was examined using fluorescence quantitative RT-PCR(FQ-RT-PCR). Results The talin mRNA level ( 126 535 copy/μg RNA) in colorectal carcinoma tissues decreased more significantly than that in adjacent normal tissues( 22 689 376 copy/μg RNA) ( P 〈 0.01 ). The carcinoma with lymph node metastasis had less talin mRNA expression than those without lymph node metastasis (P 〈 0.01 ). The deeper extent of infiltrating, the lower talin mRNA expression ( P 〈 0.01 ). Conclusions Talin mRNA shows reduced expression in colorectal carcinoma. Talin is relatod to the infiltrating and metastasis of colorectal carcinoma and might be a valuable marker for the carcinogenesis and progression of the carcinoma.
