在模拟人体生理条件(pH=7.4)下采用荧光光谱和紫外可见-吸收光谱研究了地塞米松(Dexamethasone)与牛血清白蛋白(BSA)的相互作用。荧光光谱实验结果表明地塞米松对BSA的荧光有静态猝灭作用。本工作还研究考察了298K、302K和306K三个温度下地塞米松对BSA的荧光光谱,计算了热力学常数。猝灭常数分别为1.74×106、1.62×106和1.38×106L·mol-1,结合常数分别为3.36×106、3.28×106和3.21×106L·mol-1,结合位点数分别为0.81、0.71和0.76。确定了作用力为静电引力。依据Frster非辐射能量转移理论计算了授体-受体间的结合距离。
The interaction between dexamethasone (DEX) and bovine serum albumin (BSA) was investigated by fluorescence spectroscopy in physiological buffer of pH=7.4.When DEX was added into the solution of BSA,the fluorescence intensity of BSA decreased with the increasing concentration of the added drug,indicating their interaction was involving fluorescence quenching.The Stern-Volmer quenching constant KSV,binding constant K and corresponding thermodynamic parameters of ΔH,ΔG,and ΔS were evaluated.The main sorts of acting force between the drug and BSA was found to be electrostatic force.The distances,r,between donor (BSA) and acceptor (DEX) was calculated according to fluorescence resonance energy transfer (FRET).
