目的:探讨氧化型低密度脂蛋白(oxLDL)能否诱导成熟的巨噬细胞转化为树突状细胞。方法:小鼠巨噬细胞系RAW264.7经oxLDL干预后,相差显微镜动态观察细胞形态变化,流式细胞仪检测细胞表面标志物。结果:oxLDL诱导48h后RAW264.7细胞发生树突样细胞形态改变,并表达与树突状细胞免疫成熟和激活有关的细胞表面标志物,CD40、CD86、CD83、MHC Class Ⅱ和CD1d分别上调约70%、90%、88%、76%和67%。oxLDL的作用具有剂量依赖性和时间依赖性,在10μg/ml浓度下诱导48h最佳。结论:oxLDL能够诱导成熟的巨噬细胞分化为树突状细胞,可能加剧动脉粥样硬化炎症和免疫反应。
Objective:To investigate whether mature macrophage could differentiate into dendritic cell at presence of oxidized low-density lipoprotein (oxLDL). Methods:OxLDL-treated RAW264.7 murine macrophage cell line was observed by phase contrast microscope and cell surface markers was analyzed by FACS. Results: OxLDL-treated RAW264.7 murine macrophage cell line differentiated into dendritic like cells after 48h. Cell surface markers involved in dendritic cell immune maturation and activation increased significantly. CD40,CD86,CD83, MHC Class II and CD1 d was upregulated 70 %, 90%, 88 %, 76% and 67 % respectively. The effect of oxLDL was dose dependent and time dependent; And the best effect was got at presence of oxLDL (10μg/ml) for 48 h. Conclusion:OxLDL promotes dendritic like cells transition from mature macrophage, which may aggravate inflammation and immunologic reaction of atherosclerosis.
