目的观察淫羊藿苷对自然衰老大鼠海马组织BDNF和TrkBmRNA表达的影响。方法自然衰老大鼠(24月龄)15只,随机分为对照组、淫羊藿苷低剂量和高剂量组,分别灌胃给予蒸馏水(1mL·kg^-1)和淫羊藿苷30mg·kg^-1、60mg·kg^-1,连续给药60d。观察海马组织形态学变化;实时荧光定量RT—PCR法检测各组大鼠海马组织中BDNF、TrkB mRNA的表达。结果HE染色显示对照组老年大鼠海马神经元稀疏,排列紊乱,并散在变性的神经元。给予淫羊藿苷后海马神经元排列较整齐,变性神经元减少,海马组织BDNF和TrkBmRNA表达量(306±59和389±80)与对照组(61±5和97±25)比较明显升高(P〈0.05)。结论淫羊藿苷抗衰老作用一定程度上是通过上调海马组织中BDNF和TrkB mRNA表达实现的。
AIM To investigate the effects of icariin (ICA) on the expression of BDNF and TrkB mRNA in the hippocampus of natural aging rats. METHODS Fifteen 24- month- old rats were randomly divided into control group and ICA low dose (30 mg·kg^-1) and high dose (60mg·kg^-1) groups, ig treated with distilled water and ICA respectively for 60 days. The morphological changes of hippoeampal neurons were observed by HE staining and the mRNA levels of BDNF and TrkB were detected by real-time fluorescent quantitative RT-PCR. RESULTS HE staining indicated that the number of neurons reduced and the neuronal arrangement was disordered with neuronal degeneration in the natural aging rats. ICA treatment could improve the arrangement of neurons and attenuate the neuronal degeneration. Compared with the control group, ICA apparently increased the expression of BDNF and TrkB mRNA in the hippoeampus of aging rats. CONCLUSION The anti-aging effects of ICA might be related to the increased BDNF and TrkB mRNA expressions in the hippoeampus.
