人二倍体细胞对不同病毒感染过程的生物学效应比较分析

目的分析人二倍体细胞对不同病毒感染的生物学反应差异性及相关分子生物学机制。方法用人类肠道病毒71型(enterovirus 71,EV71)和人单纯疱疹病毒1型(herpes simplex virus type 1,HSV-1)感染人二倍体细胞,光镜和电镜下观察感染细胞形态学特征;比较两种病毒在人二倍体细胞上的增殖动力学及两种病毒感染后细胞酶谱的变化;采用基因芯片检测两种病毒感染后人二倍体细胞基因反应的差异。结果感染细胞的光镜或电镜的超微结构观察均表现出许多相同的特点,包括细胞变圆、肿胀、折光性增强,以及细胞细微结构的受损,大多数细胞器的破坏等。两种病毒在人二倍体细胞上具有不同滴度峰值的增殖动力学过程;对细胞超氧化物歧化酶的动力学改变趋势有不同影响,但对乳酸脱氢酶和琥珀酸脱氢酶动力学变化趋势无明显影响;两种病毒感染引起人二倍体细胞出现不同的基因反应,其中与信号转导及与天然免疫反应有关的mRNA转录效率具有一定的差异。结论人二倍体细胞对EV71、HSV-1感染过程表现不同的生物学效应。

英文摘要:

Objective To analyze the differences of the biological responses of human diploid cells to various virus infection as well as the relevant molecular mechanism. Methods Human diploid cells were infected with human enterovirus 71(EV71) and human herpes simplex virus type 1(HSV-1) respectively, and observed for morphological characteristics by light and electron microscopies. The proliferation kinetics of two viruses in human diploid cells and the changes of cell enzyme spectrum were compared, and the difference of gene responses of infected cells was evaluated by DNA microarray. Results The ultrastructures of infected cells under light or electron microscope showed identical characters including rounding, swelling and increased refractivity of cells, as well as damages of microscopic structure and most of cell organelles. Different peak values of titers were observed in the proliferation kinetics of two viruses in human diploid cells, which showed different effect on the change tendency of kinetics of cell superoxide dismutase, while showed no significant effect on those of lactate dehydrogenase and succinate dehydrogenase. The infection with either of the viruses caused different gene responses in human diploid cells, including a certain difference in m RNA transcription efficiency related to signal transduction and natural immune response. Conclusion Human diploid cells showed different biological effects during infections with EV71 and HSV-1.
