目的:研究热毒宁注射液雾化吸入的刺激性。方法:健康SD大鼠12只,随机分为生理盐水组、热毒宁低浓度组、热毒宁高浓度组,每组4只,各组每日给药2次,每次雾化吸入20min,连续给药7d,观察大鼠行为变化,于第8天取口腔黏膜、鼻中隔黏膜、气管、左肺,制备石蜡切片,HE染色,观察形态学结构变化。同时,右肺做肺泡灌洗术,取灌洗液离心并测定蛋白浓度及乳酸脱氢酶(LDH)活力。结果:大鼠有不同程度的行为反应,给药组有鼻腔出血和眼部出血的现象。切片观察可知热毒宁对黏膜有一定刺激性,且热毒宁高浓度组刺激性大于热毒宁低浓度组。3组肺泡灌洗液蛋白浓度均无显著性差异,而LDH活力随热毒宁浓度的增大而显著增大(P〈0.05)。结论:热毒宁雾化吸入给药方式的刺激性较明显,临床应用须谨慎。
Objective: To study the irritation of Reduning Injection aerosol inhalation from the views of animal behavior, morphology changes, protein oncentration and lactate dehydrogenase(LDH) activity. Methods: Twelve male adult rats were randomly divided into 3 groups: the normal saline group, low concentration Reduning group, high concentration Reduning group, 4 rats per each group. The Reduning was given by inhalation administration for 20 minutes each time, twice a day, for 7 days. The rat behavior was observed during the experiment. Then, to observe the morphology changes, the mouth mucosa, nasal septum mucosa, trachea and left lung tissues were harvested to prepare the paraffin sections for HE staining. The bronchoalveolar lavage operation was used to determine the protein concentration and LDH vitality. Results: The bleeding in nasal cavity and eyes could be obviously seen in the treatment groups as well as some behavior reactions. The pathological results showed that Reduning had irritant effects on the tissue mucosa, particularly in the high concentration Reduning group. There was no difference in protein concentration among 3 groups, however, LDH was more active depended on concentration increasing(P〈0.05). Conclusion: Reduning aerosol inhalation may irritate mucosa, so it should be used with extreme caution.
