Microbial degradation has been suggested as the best way to remove polycyclic aromatic hydrocarbons (PAHs) from contaminated environment, in this study, two naphthalene-degrading mixed cultures H1 and H2 were enriched from the petroleum-contaminated soil around a gas station, and one preponderant pure bacterial strain C1 utilizing naphthalene as a sole carbon and energy source was isolated from the mixed cultures. The results indicate that culture medium with naphthalene as a sole carbon and energy source is better than with other carbon nutriment in screening microorganism capable to degrade naphthalene. It was found that after 40h incubation in mineral salt medium with an initial naphthalene concentration of 1000 mg·L^-1, the cell numbers of H1, H2 and C1 had increased more than ten times and the degrading rates of naphthalene were over 99.6%.
