目的研究粉防己碱(Tet)对神经细胞兴奋性氨基酸N-甲基-D-天冬氨酸(NMDA)损伤的保护作用。方法采用原代培养的大鼠皮层神经细胞模型,给予NMDA直接损伤,加入不同浓度的粉防己碱,测定神经细胞损伤MTT变化。结果粉防己碱对兴奋性氨基酸损伤保护作用在10^-8~10^-6mmol/L范围内存在非剂量线性相关。结论Tet通过多靶点对神经元兴奋损伤产生拮抗作用,离体细胞模型中5×10^-7mol/L浓度具有最佳的保护效果。
Objective To investigate the neuroprotective effect of Tet for neurons damaged by NMDA. Methods By using primary cultured rat cortical neurons, the changes of neurons in MTF damaged by NMDA and protective effects of Tet were observed. Results The neuroprotective effects of Tet is closely related to the concentration of Tet at 10 ^-8 ~ 10^ -6 mmol/L and they were unrelated to its linear dosage. Conclusion The neuroprotective effects of Tet are operated in universality, and 5 × 10^ -7mmol/L is the best concentration for its neuroprotective effect.
