光核桃是我国特有的蔷薇科桃属植物,具有较强的耐旱、抗病能力和更新容易等特性。提取高质量的基因组DNA是研究光核桃的分子特性的关键所在。为了获得高质量的光核桃基因组DNA,利用CTAB提取法对其提取方法进行了优化。结果表明:CTAB提取液的最佳用量为700μL,最佳裂解时间为40 min,酚、氯仿、异戊醇混合液(体积比为25∶24∶1)的最佳使用次数是2次,氯仿、异戊醇混合液(体积比为24∶1)的最佳使用次数也是2次,异丙醇的最佳使用体积分数为2/3。
Amygdalus mira belongs to Rosaceae and Amygdabus from China,and has strong drought tolerance,disease-resistant,easy to update,and other characteristics.The key to researching molecular properties in A.mira is to get high-quality genomic DNA.In order for reaching the goal,the CTAB extraction method was optimized.The results show that the best dosage of CTAB extracts is 700 μL,the best incubation time is 40 min,the best usage frequency of the mixture of phenol,chloroform and isoamyl alcohol(25∶24∶1) is two times,the best usage frequency of the mixture of chloroform and isoamyl alcohol(24∶1) is also two times,and the best volume ratio of isopropyl alcohol is 2/3.
