玻璃化被认为是低温保存关节软骨的一种有效方法,但需要考虑低温保护剂渗入软骨的速率以及低温保护剂毒性的制约。本文选用二甲亚砜(DMSO)作为低温保护剂,对其在1℃,22℃和37℃时对绵羊关节软骨的渗透特性进行了研究,得到相应的扩散系数为1.9×10^-6 cm^2/s,3.3×10^-6 cm^2/s和5.7×10^-6cm^2/s,活化能为22.84 kJ/mol。认识低温保护剂对关节软骨的渗透特性有助于建立低温保护剂加入/取出过程的数学模型,从而优化关节软骨的玻璃化保存方案。
Vitrification is considered to be a promising method for cryopreservation of articular cartilage,but the permeation rate of cryoprotectant(CPA) into the matrix and the toxicity of CPA should be considered.This study examined the permeation of dimethyl sulfoxide(DMSO) into ovine articular cartilage at 1℃,22℃and 37℃.The resulting diffusion coefficients are 1.9×10-6 cm2/s, 3.3×10-6 cm2/s and 5.7×10-6 cm2/s respectively,and the activation energy is 22.84 kJ/mol.These data are helpful to build the mathematical model of CPA addition and removal,and consequently optimize the vitrification protocol of articular cartilage.
