目的探讨凝血酶诱导的胎鼠大脑皮层神经元损伤与RhoA蛋白活化表达的关系。方法体外培养胎鼠大脑皮层神经元8d后,采用Western blot技术检测不同浓度凝血酶(0、1、10、30和100U/m1)组作用3h后及30U/ml凝血酶作用不同时间(O、0.5、1、3和6h)组胎鼠大脑皮层神经元RhoA蛋白表达。结果凝血酶30U/ml组神经元细胞膜RhoA蛋白表达高于0U/ml组(P〈0.05);5个凝血酶浓度组RhoA总蛋白表达量无统计学差异。凝血酶30U/ml作用3h组细胞膜RhoA蛋白表达高于0h组(P〈0.05);5个时间组RhoA总蛋白表达量无统计学差异。结论凝血酶诱导的胎鼠大脑皮层神经元损伤与一定浓度凝血酶和一定作用时间后RhoA蛋白的激活有关。
Objective To investigate the relationship between the activation of RhoA protein and thrombin-induced cortex neuron injury in fetal rats. Methods The fetal rat cortex neurons were cultured in vitro for 8 days, which then were divided into 5 groups of different thrombin concentrations (0,1,10,30 and 100 U/ml) and acted for 3 hours, and 5 groups of different times(0,0. 5,1,3 and 6 h) for thrombin 30 U/ml to act. The expressions of RhoA protein were detected by Western blot and compared among the ,groups. Results The expression of RhoA protein on cell membrane was significantly higher in thrombin 30 U/ml group than that in thrombin 0 U/ml group(P〈0. 05). The total expression of RhoA protein was not significantly different among 5 groups of different thrombin concentrations. The expression of RhoA protein on cell membrane was significantly higher in group of thrombin 30 U/ml acting for 3 hours than that in group of thrombin 30 U/ml acting for 0 hour (P〈0. 05). The total expression of RhoA protein was not significantly different among 5 groups of thrombin 30 U/ml acting for different times. Conclusion The cortex neuron injury induced by thrombin in fetal rats is related to the activation of RhoA protein in a certain concentration of thrombin and a certain time of action.
