天然免疫受体在机体抗病毒过程中发挥着重要作用。为了解Nalp3、MDA5和TLR7受体在青年鸡组织中分布特征,首先应用RT-PCR方法从18周龄鸡肝脏中分别扩增目的片段,建立实时荧光相对定量RT-PCR检测多种组织中Nalp3、MDA5和TLR7受体的表达谱及相对含量。结果表明,3种受体在不同组织中转录水平差异较大,其中Nalp3在肝脏和肾脏、TLR7在心、脾和肺脏中表达量相对较高,而MDA5在各组织中表达均较低。该3种受体在不同组织中转录水平的差异提示其识别的相对作用可能不同。
Innate immune receptor plays a major role in antiviral responses. Here,distributions of transcription of the patternrecognition receptors Nalp3,MDA5 and TLR7 in tissues were analyzed.The gene fragments of three receptor genes were amplifiedfrom young chicken livers with the specific primers. The quantitative real-time reverse transcription PCR(RT-q PCR)was estab-lished for the detection of the receptor m RNAS in different tissues of young chicken at the age of 18 weeks.The results showed thatthere were comparatively large differences in the transcription levels of three receptor m RNAS in different tissues. Nalp3 gene ex-pression in the liver and kidney was relatively higher,and TLR7 expression in the heart,spleen and lung was relatively higher.However,MDA5 expression was very low in different tissues.Difference transcription levels of the three receptors may indicate thattheir differences reflect their relative roles in chicken.
