卵母细胞的异位移植技术能明显提高干细胞来源的卵母细胞的发生效率和质量,而原位移植技术目前国内外报道很少。本实验旨在建立原位移植诱导卵母细胞发生的模型,为后续的干细胞向卵母细胞的定向分化提供技术基础。对成年小鼠使用白消安处理后,其卵巢内的卵泡大量减少,4周时卵巢内卵泡的数量最少。利用白消安处理4周的小鼠作为受体,向其卵巢内注射EGFP转基因胎鼠原始生殖细胞。注射30天后,在受体小鼠卵巢中可以发现EGFP标记的原始生殖细胞来源的卵母细胞参与卵泡的形成与发育。本研究证明,通过原位移植技术,原始生殖细胞在成年小鼠卵巢中分化成卵母细胞,并参与卵泡的形成与发育。
Ectopic transplantation technique can significantly improve the efficiency and quality of stem cell - derived oocytes, whereas ovarian orthotopic transplatation was rarely reported both at home and abroad. The aim of the present study is to establish the model of oogenesis system induced by intraovarian transplantation, which would provide a technical basis for our follow - up study of inducing stem cells into the oocytes. After busulfan treatment, adult female recipients mice underwent significant decrease of growing follicles and reduced to a minimum at 4 weeks after treatment. So EGFP labeled primordial germ cells (PGCs} were purified from 12.5dpc fetal ovaries, and intraovarian transplanted into the ovaries of recipients mice 4 weeks after treatment. 30 days after transplantation, immunofluorescence revealed that EGFP possitive oocytes were involved in the formation of growing follicles. In conclusion, after intraovarian transplantation, PGCs are differentiated into oocytes and are involved in folliculo- genesis in adult ovaries.
