目的:观察健脾补土法组方对脑缺血再灌注(I/R)大鼠脑组织基质金属蛋白酶(MMP)-2的表达及血脑屏障通透性的影响。方法:将50只大鼠随机分为假手术组、模型组、健脾补土小剂量组、健脾补土大剂量组、依达拉奉组,除假手术组外,其余组小鼠均运用大脑中动脉栓塞再通法建立脑I/R模型。在造模前3d用相应药物对各组小鼠进行灌胃或腹腔注射。造模完成后对其进行神经功能评分、检测血脑屏障通透性及脑组织MMP-2的表达。结果:与模型组比较,健脾补土法各组治疗大鼠的神经功能评分均有改善,血脑屏障通透性降低,MMP-2的表达减少(P〈0.05,P〈0.01)。结论:健脾补土法保护血脑屏障可能与其抑制MMP-2表达有关。
Objective: To observe the effects of spleen -strengthening therapy on the matrix metalloproteinase (MMP) -2 expres- sion in brain tissue and blood -brain barrier permeability in rats with cerebral ischemia/reperfusion (I/R) injury. Methods: Fifty rats were randomly divided into sham operation group, model group, low - dose spleen - strengthening therapy group, high - dose spleen - strengthening therapy group, and edaravone group. In addition to sham operation group, the rest of the rats were induced cerebral IfR models by middle cerebral artery occlusion and recanalization. Medicines were given to the rats by garage or intraperitoneal injection at 3 d before the cerebral I/R model was induced. The neurological scores were evaluated; the blood - brain barrier permeability and MMP - 2 expression in the brain tissue were measured. Results : Compared with the model group, the low - dose spleen - strengthening therapy group and high - dose spleen - strengthening therapy group had significantly improved neurological scores and significantly de- creased blood - brain barrier permeability and MMP - 2 expression ( P 〈 0.05 or P 〈 0.01 ). Conclusion : Spleen - strengthening thera- py can protect the blood -brain barrier, which may be due to its inhibitory effect on MMP -2 expression.
